       @escarpment @futurebird what I'm saying in a roundabout way is that we have a single origin in part because the software we use to generate visualisations assumes a single origin.

       @escarpment @futurebird no, not a metaphor at all.Look at any phylogenetic tree in any publication.        @escarpment @futurebird yes, as far as we know, DNA is the only thing that creates life, as we understand it.However...1. There is tentative link to the creation of a nucleic acid or amino acid world from "nothing", so it's not completely out of the realm of possibility to suggest that there were multiple origins that were spliced together.2. Nature has other non-DNA ways to replicate (fire is my favourite example), but we don't treat the things that replicate in a non-DNA as being alive.

       @nanopore It seems the reason I'm getting confused about the precise sequences in the TrackIt 1kb Plus DNA ladder is that there are multiple different sequences that fit into the same band length.Either that, or they're fragments of longer bands that have been clipped at very specific lengths.#DNAladder #Flongle #LSK114

       @nanopore I'm up to 650bp now.Despite the longest sequence so far having a reported BLAST identity of 98%, I've double-checked the reads, and decided that it's more likely that there's variation from the closest matching reference than a sequencing error.#DNAladder

       @nanopore 1kb seems to be about the limit of what I can pick out easily with LSK114 + Flongle.But I think that's probably okay, because from memory the longer ones were easier to work out / assemble from RBK-fragmented reads.Then again.... maybe I should just go all out and use a PromethION flow cell with LSK114.
