Greetings Bionetters,

I was wondering if anyone out there has had any expereince in doing PCR on
insect leg material.

I'm going to be starting to screen insects using PCR and want to be able to
identify individuals carrying specific sequences without killing them.

I was planning on pulling a single leg from individuals and pooling say 50-100
legs, homogenizating them and then doing a crude purification before the PCR.
The fewer steps needed the better. The PCR will be needed to detect a single
copy sequence present in at most 1 individual per pool.

Any suggestions/expereinces/references most appreciated.
