We would like to make the separation of our YACs away from yeast chromosomes easier by digesting away yeast chromosomes with MluI before pulse-field separation. Has anyone tried this strategy or something like this? Is anyone else out there trying to transform with YACs? How successful have you been? ~~~~~~~~~~~~~~~~~~~~~~~~~~~~ ~ ~ - - ~ David Fitch ~ \ / / / ~ Department of Biology ~ \ / / ~ New York University ~ [] / / ~ Main Building, Room 1009 ~ \ / / ~ New York, NY 10003 ~ \ / ~ 212-998-8254 ~ \ / ~ fax 212-995-4015 ~ / ~ fitch@acf2.nyu.edu ~ / ~~~~~~~~~~~~~~~~~~~~~~~~~~~~ .
