For example, a 20 base-pair sequence, but the middle of one strand is not connected, what will be the melting behavior? Any similar experimental or simulation results available? G-G-G-G-G-G-G-G-G-G-A-A-A-A-A-A-A-A-A-A C-C-C-C-C-C-C-C-C-C T-T-T-T-T-T-T-T-T-T As mentioned before, 20 base-pair sequence, but there is no connection between the last base C and the 1st base T. Presumely, the melting will be determined by the 1st half of the sequence, since it's more stable, but will it be possible to have two peaks on the derivative of the melting curve? Any comments or reference will be appreciated.
