The rearrangement has been seen w. r. t. an X-chromosome specific repetitive DNA probe. So, I need to do Southern hybridization of embryonic vs. salivary gland DNA(from third instar larvae), with this probe, to be comletely sure of the rearrangement. Ashburner's lab manual (referring Cohen and Gotchell, 1971)gives a method using a specific kind of pestle and a very finely roughened homogenizing tube. It is impossible for me to get anything, with that kind of specifications, made here. If anyone has seen any other method, can write the method to me or let me know the reference. Otherwise any unpublished, working protocol will also be of great help and will be duely acknowledged. If anyone can donate me that special kind of pestle, used by Cohen and Gotchell, it will be highly appreciated. Thanking you in advance for any kind of help.
