Both are by James S. Clegg: Am. J. Physiol. 246 (Regulatory Integrative Comp. Physiol. 15): R133-R151, 1984, Properties and metabolism of the aqueous cytoplasm and its boundaries and Journal of Cell Biology, 99:167s-171s, 1984, Intracellular water and the cytomatrix: some methods of study and current views The main take-home that I got from these is that one should expect purified enzymes to behave quite differently from in-vivo, and that even microsomal preps (my bread and butter for the past few years) or "cytosol" are suspect. Since I'm interested in metabolism and distribution in mammals (including humans) this is a problematic issue. For the liver, freshly isolated hepatocytes are then preferred, but there is a fair bit of art/effort to getting and using them on a regular basis, when sub-cellular preps are sooooo much easier. sigh Paul S. ~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~~ Paul M.
