I'd like to remove inserts from the Okkema lambda gt11 library using primers flanking the EcoRI site in the Bgal coding region, and am having difficulty getting PCR products this way (i.e. by taking 1 uL of supernatant from a lambda plaque). I'd appreciate any comments or suggestions.
