We are not C. elegans people. But we are excited by the development in C. elegans field. So we would like to borrow some new tricks you guys did to C. elegans and try to use them on plant parasitic nematodes we are interested in. First we want to microinject DNA into the female gonad to transform them. But these females have thick cuticles and they are not transparent at all! So we are now thinking to transform the males and let the males do the job instead. But we are not sure whether after injecting DNA into male gonad, we will end up with transformed progenies. So help wanted!!! This may sound silly! BUT: 1. Anybody ever tried microinjection in males of C. elegans? 2. Can you see the male gonad as clear as hermaphrodite gonad? 3. If I can put DNA into male gonad, do you think the foreign DNA will go together with sperms to fertilize the oocytes? Will the cytoplasm volume of oocyte or sperm decide the chance of trnaformation? Will the foreign DNA interact with all those nucleus in male gonad, hence they get incorporated into sperms? Your suggestion will be highly appreciated! HELP WANTED! Thank you! .
