It's my understanding that the nuclei are very fragile and as such are hard to isolate intact. On the other hand, can anyone see a problem with using unfractionated cellular protein for gel shift experiments? Also, how much sucess have peple had at doing immunolocalization on polytene chromosomes? (i.e. looking at DNA binding proteins)? Any feedback will be appreciated. Thanks!
