I am trying to find a protocol which will keep the cells attached to slides as they undergo a number of washes and incubations in preparation for viewing with immunofluorescence microscopy. So far I have tried coating my slides with Poly-L-Lysine, Biobond and 3-aminopropyl- triethoxysilane (APTES). Of these agents, APTES produced the greatest cell retention although I still lost most of the original cells. Any advice will be greatly appreciated.
