If you were replacing or update a shared object file (like DLL's) that is being used by an app you would probably need to exit that app and then restart it. The kernel is pretty much self contained. It has to be since the partition with the library files won't be mounted or available when the kernel is starting up. If you were replacing or updating part of KDE or Gnome, you would just need to restart that part. No need for a reboot.
